Melatonin Synthesis: An Overview

My naturopath recommended 5-htp, which I had taken years ago and not noticed much effect, and also took it for a bit while taking 5-htp, and found that it helped with sleep onset.
She started me on 100mg.

What is the difference between Melanin and Melatonin

Dieting in healthy women results in a decrease in the availability of tryptophan, the amino-acid precursor of serotonin, for brain serotonin synthesis.

In addition, the antiaromatase and antisulfataseeffects of melatonin have also been shown in cancer cell typesother than breast cancer cells. In glioblastoma cells, whichexpress estrogen receptors and have the ability to synthesizeestrogens, melatonin also reduces the local production of estrogensby decreasing the activity of aromatase, sulfatase and 17β-HSD1,and downregulating aromatase, sulfatase and 17β-HSD1 mRNA steadystate levels (,).

Serotonin is not sold since it cannot easily cross the blood brain barrier while tryptophan, 5-HTP and melatonin easily cross from the blood stream into the brain.

Biochemical pathway of melatonin synthesis in ..

is converted to melatonin through the sequential action of two enzymes, serotonin N-acetyltransferase (arylalkylamine N-acetyltransferase, or AANAT) and hydroxyindole-Omethyltransferase (HIOMT).

Melatonin is synthesized using a four step pathway from it’s precursor: Tryptophan

The increased synthesis of melatonin caused by norepinephrine ..

Gonadotropin-inhibitory hormone (GnIH), a neuropeptide that inhibits gonadotropin synthesis and release, was first identified in the quail hypothalamus. To understand the physiological role of GnIH, this review will demonstrate the mechanisms that regulate GnIH synthesis and release. Pinealectomy (Px) combined with orbital enucleation (Ex) decreased the synthesis of GnIH precursor mRNA and content of mature GnIH peptide in the diencephalon. Melatonin administration to Px plus Ex birds caused a dose-dependent increase in the synthesis of GnIH precursor mRNA and production of mature peptide. A melatonin receptor subtype, Mel1c, was expressed in GnIH-immunoreactive neurons, suggesting direct action of melatonin on GnIH neurons. Melatonin administration further increased GnIH release in a dose-dependent manner from hypothalamic explants in vitro. GnIH mRNA expression and GnIH release during the dark period were greater than those during the light period in explants from quail exposed to long-day photoperiods. Conversely, plasma luteinizing hormone (LH) concentration decreased during the dark period. This review summarizes that melatonin appears to act on GnIH neurons in stimulating not only GnIH synthesis but also its release, thus inhibiting plasma LH concentration in birds.

Tryptophan is also a precursor to the neurotransmitter serotonin and the hormone melatonin ..

This melatonin modulatory effect on the aromataseand sulfatase enzymes, at the tumor cell level, has also beendescribed , in rats bearing7,12-dimethylbenzanthracene-induced mammary tumors. The growth ofthese mammary tumors is estrogen-dependent and ovariectomysignificantly reduces both the size and number of tumors, while theadministration of testosterone or estrone sulfate to ovariectomizedanimals is able to maintain the tumor growth at the same level asthe control (uncastrated) animals. The stimulatory effects of tumordevelopment induced by testosterone (which depends on the localsynthesis of estrogens from androgens), due to the aromataseaction, or estrone sulfate (which depends on the estrogens locallyformed by the action of the sulfatase enzyme on the biologicallyinactive estrogens), are suppressed by the administration ofmelatonin. Tumors from animals treated with melatonin have thelowest microsomal aromatase and sulfatase activity (,).
