It is possible to determine rates of CO2 assimilation and water loss (transpiration) by measuring the flux of CO2 and water vapour from a leaf in a sealed chamber. This process, termed gas exchange (because CO2 is going in and water vapour is coming out) is more complicated than might be initially imagined. During photosynthesis, plants take up CO2 (which is converted to sugar) and produce oxygen. All the while they are respiring and releasing CO2 back into the cells. To make matters more complex, the enzyme that fixes CO2 (Rubisco) also ‘fixes’ oxygen, a reaction called photorespiration that releases CO2, but does not produce energy. Gas exchange is also influenced by light levels, because when more light is available, generally, more CO2 can be fixed.

Physiological factors that affect photosynthetic rates

Our results indicated that photosynthesis occurred fastest at 100 watts for both plants. This was inconsistent with our hypothesis. There were several possible problems with our experiment. We only ran one trial for each plant under the various light intensities due to time constraints. This made it impossible for us to run a statistical test. If more trials were available, we could have run a linear regression analysis to see if there was a significant linear correlation between intensity of light and rate of photosynthesis. Another problem is that 100 watts may have not been intense enough. The slope of the shade plant seemed to be decreasing as intensity increased, and the slope of the sun tolerant plant seemed to be increasing. If we had used higher intensities of light, perhaps these trends in the slope would have continued and the shade plant's slope would have started to decrease, allowing us to fail to reject our hypothesis. The spike in photosynthesis for the shade tolerant plant could indicate that they are not accustomed to sudden changes in light intensity. The slow rise in the photosynthesis rate for the sun tolerant plant could indicate that it is more equipped to maintain homeostasis in sudden intensity changes.
